Documentation and traceability for reference material

The purpose of these records is narrow and worth stating plainly: being able to tell, later, whether an unexpected result came from the experiment or from the material.

The inventory record

Per lot, not per compound: compound name and identifier, lot number, supplier, receipt date, nominal quantity, net peptide content if reported, storage location, condition on arrival, and a link to the certificate file.

Keying on lot rather than compound is the part that does the work. A compound-level record cannot answer the question that actually gets asked — did the results change when we opened the new vial — because it has already merged the lots together.

Recording net peptide content at intake rather than looking it up at preparation time removes a step at the moment when someone is most likely to skip it.

The preparation record

When a vial is reconstituted, record the lot, the solvent and its concentration or buffer composition, the volume added, the resulting concentration, whether that concentration is nominal or corrected for net peptide content, the date, and who prepared it.

The nominal-versus-corrected flag deserves emphasis. A concentration written without it is ambiguous, and the ambiguity is invisible — the number looks equally authoritative either way. Two people can then compare results computed on different bases without either realising.

Aliquots should carry enough to be identified standalone: compound, lot, concentration, solvent, date. A tube labelled only with a compound name is a tube of unknown concentration as soon as it leaves the rack it was prepared in.

Linking data to material

Experimental records should reference the lot, not just the compound. This is the link that makes retrospective analysis possible, and it is the one most often omitted because at the time of the experiment there is only one lot and the distinction feels pedantic.

It stops feeling pedantic the first time results shift after a restock. With lot references in the data, the correlation is visible immediately. Without them, the investigation starts from nothing and often concludes with an unsatisfying shrug.

Freeze-thaw count per aliquot is worth tracking where solutions are stored rather than prepared fresh, since cumulative cycling is a plausible explanation for gradual drift that no single measurement will reveal.

Retention

Keep certificates for as long as any data derived from that lot is in use — which in practice means considerably longer than the material itself lasts. A certificate discarded when a vial empties leaves the resulting data unsupported.

Storing certificate files alongside the inventory record, rather than in a separate document store organised by supplier or date, is what keeps them findable at the moment someone actually needs one.

Common questions

Why record lot numbers in experimental data?
It is the only way to later correlate a change in results with a change in material. Without it, an investigation into a shifted baseline has no way to test the most likely explanation.
What is the most commonly omitted field?
Whether a recorded concentration was corrected for net peptide content. The number looks equally valid either way, so the ambiguity is invisible until two people compare results computed differently.
